一、意思不同

1、a number of

意思是：许多，一些

例如：A number of students are sick today.

今天一些学生病了。

这里 a number of 相当于  many 或者some。

2、the number of

意思是 ......的数量

The number of sick students is decreasing.

生病学生的数量在减少。

二、中心词不同

1、the number of中心词是number

例如：The number of students in our class is 50.

我们班学生的数量是50人。

2、a number of，number前可用large、small等修饰以表示程度。

例如：A number of students are planting trees on the hill.

许多学生在小山上植树.

三、谓语动词使用形式不同

1、the number of谓语动词要用单数。

2、a number of作主语时,谓语动词用复数。

为什么number这个单词在句中第一个字母要大写？

因为在英语中，表示几年级，几班，几号，几室是需要大写的，例如三年二班是 Class2, Grade 3；5号是Number5, 一般缩写为No.5；501室是Room 501。所以number这个单词在表示号码的时候时，，在句子中需要大写。
